Use of novel cystine analogs to decrease oxidative stress and control product quality

Continuous improvements of cell culture media are required in order to ensure high yield and product quality. However, some components can be instable lead detrimental effects on bioprocess performances. l-cysteine is an essential amino acid commonly used media. Despite its beneficial effect recombinant protein production, cases, this component responsible for microheterogeneity. In context, alternative have found reduce variants while maintaining productivity. study, we assessed the performance different cysteine cystine analogs : N-acetyl-cysteine, s-sulfocysteine, N,N′-diacetyl-l-cystine dimethylester (DACDM). Replacement by analogs, especially DACDM, has shown positive impact charge level coloration level. Moreover, molecule contributed increase intracellular glutathione pool, which suggests a close relationship with oxidative stress regulation.
